Enhanced micronucleus test in human samples through automated micronuclei segmentation

Biological monitoring to ensure the health and safety of workers exposed to carcinogens and mutagens is required by Italian law. Among the various biomarkers available, micronucleus test is effective in individuating DNA damage and predicting carcinogenesis. However, the decree lacks specific guidelines on how these tests should be implemented, leading to their use primarily within scientific research settings.
This work proposes an innovative approach to develop a non-invasive, quick, and cost-effective method for conducting the micronucleus test. The test is performed on immature erythrocytes (reticulocytes) from human blood smears using a specialized automated software. This software is designed to automatically identify reticulocytes based on morphometric (shape and size) and colorimetric (presence of orange color) parameters, and detect any micronuclei present. 
It facilitates the automated image acquisition from blood smears and will be validated with positive controls.
Implementing this system could significantly improve worker health and safety by enabling the early detection of damage and allowing for timely intervention with corrective or protective measures to prevent the onset of occupational diseases.
